Brain cholinesterase activity of apparently normal wild birds

Organophosphorus and carbamate pesticides are potent anticholinesterase substances that have killed large numbers of wild birds of various species. Cause of death is diagnosed by demonstration of depressed brain cholinesterase (ChE) activity in combination with chemical detection of anticholinesterase residue in the affected specimen. ChE depression is determined by comparison of the affected specimen to normal ChE activity for a sample of control specimens of the same species, but timely procurement of controls is not always possible. Therefore, a reference file of normal whole brain ChE activity is provided for 48 species of wild birds from North America representing 11 orders and 23 families for use as emergency substitutes in diagnosis of anticholinesterase poisoning. The ChE values, based on 83 sets of wild control specimens from across the United States, are reproducible provided the described procedures are duplicated. Overall, whole brain ChE activity varied nearly three-fold among the 48 species represented, but it was usually similar for closely related species. However, some species were statistically separable in most families and some species of the same genus differed as much as 50%.     

Preparation and properties of prostaglandin D2 labelled with tritium with high molar radioactivity

Kinetic parameters of enzymatic and non-enzymatic transformations of [3H]prostaglandin H2 (PGH2) were determined; the maximum yield of [3H]PGD2 being obtained at the keobs/koobs ratio equal to 10. The two-stage enzymatic synthesis of [3H]PGD2 with high molar radioactivity (3.15 TBq/mmol) from [3H]arachidonic acid carried out. Its identity in properties to the natural PGD2 was shown in experiments on the inhibition of ADP-induced aggregation of thrombocytes and on enzymatic oxidation with 15-hydroxyprostaglandin dehydrogenase.     

Ethanol production from pentoses by immobilized microorganisms

The capabilities of immobilized Fusarium oxysporum f. sp. lini, Mucor sp., and Saccharomyces cerevisiae in fermenting pentose to ethanol have been compared. S. cerevisiae was found to have the best fermentation rate on d-xylulose of 0.3 g l^?1 h^?1. By using a separate isomerase column for converting d-xylose to d-xylulose and a yeast column for converting d-xylulose to ethanol, an ethanol concentration of 32 g l^?1 was obtained from 10% d-xylose. The ethanol yield was calculated to be 64% of the theoretical yield.     

Assay of 1L-myo-inositol-1-phosphatase using a fluorometric method.

1L-myo-Inositol-1-phosphatase, an enzyme purified from brain tissues, catalyzes the dephosphorylation of 1L-myo-inositol 1-phosphate. This enzyme has become the subject of intense research interest, since myo-inositol is needed for the resynthesis of phosphatidylinositol. We have developed a sensitive fluorometric assay for detecting the activity of 1L-myo-inositol-1-phosphatase. The assay is based on o-aminobenzoyl beta-glycerophosphate fluorescence, according to the following principles: (I) The fluorescence yield of o-aminobenzoyl beta-glycerophosphate is increased by 2.75-fold in the presence of saturating concentrations of bovine serum albumin. (II) o-Aminobenzoyl beta-glycerophosphate has the same fluorescence yield as o-aminobenzoyl glycerol, but the latter does not bind to bovine serum albumin. (III) Dephosphorylation of the substrate, catalyzed by the monophosphatase, makes less o-aminobenzoyl beta-glycerophosphate available for binding to bovine serum albumin, thereby producing a decrease in the fluorescence intensity.

Development of neuron-capillary interrelationships in the nuclei of the anterior portion of the human hypothalamus during ontogenesis

Three periods in the formation of the spatial arrangement of the neuronal nuclei have been stated: 1) from the prenatal period, infancy including--pronounced decrease in the density of the neuronal cells arrangement; 2) from the childhood up to adolescence--relative stabilization in the hypothalamic cellular composition; 3) mature and old age--secondary decrease in the dencity of the neuronal cells arrangement. Certain peculiarities in the nuclear capillary composition and in their neuronal-capillary relationships have been revealed: during the prenatal ontogenesis, the dencity of the neuronal cells arrangement decreases and their size increases that is followed by decrease in their capillary loops. In postnatal ontogenesis no proportional relationship between the dencity of neuronal cells arrangement and that of capillary network in the nuclei of the anterior hypothalamus was noted.
